　　儀器 SPR 是基于表面等離子體共振（SPR）技術(shù)來實(shí)時(shí)跟蹤生物分子間的相互作用，不需任何標(biāo)記物的技術(shù)。該設(shè)備通過固定波長(zhǎng)，測(cè)量角度反射光譜獲得分子相互作用信息；也可固定入射角度，測(cè)量分子相互作用動(dòng)力學(xué)曲線。SPR可進(jìn)行：1、動(dòng)力學(xué)常數(shù)的測(cè)定；2、樣品濃度的測(cè)定；3、分析相互作用模式；4、復(fù)合物功能分析。

　　如圖所示，SPR 分子相互作用儀器的原理是波長(zhǎng) 632.8 nm 的激光形成的 p 偏振光，到達(dá)棱鏡后激發(fā)金屬表面的表面等離子體發(fā)生共振。通過改變?nèi)肷涔饨嵌?θ，實(shí)時(shí)檢測(cè)反射光強(qiáng)度的變化 R，根據(jù)反射光強(qiáng)度變化 ΔR 或共振角度的變化 Δθ，實(shí)現(xiàn)分子相互作用的檢測(cè)；也可以通過固定角度，測(cè)量反射光強(qiáng)的變化曲線獲得分子相互作用動(dòng)力學(xué)常數(shù)。

The detection of molecular interactions can be achieved by the changes of ΔR or Δθ; the molecular interaction kinetic constants can also be obtained by measuring the reflectivity changes at a fixed angle.
